1. What is HICA? Understanding the Principle at a Glance
HICA (Homogeneous Immuno Chemiluminescence Assay) is a liquid-phase detection technology based on energy transfer between donor and acceptor beads:
- The target antigen binds with biotinylated antibodies and chemibeads (acceptor beads), forming a sandwich complex;
- Sensibeads (donor beads) are added, and the two types of beads "meet" in the liquid phase via biotin-streptavidin binding;
- Under 680nm excitation, the donor bead's photosensitizer generates singlet oxygen, which diffuses to the acceptor bead, triggering a 612nm chemiluminescent signal;
- The signal intensity correlates with antigen concentration, enabling direct readout.
