Surface plasmon resonance(SPR) is an optical based analysis technology.SPR technology utilizes the resonance between evanescent waves and metal surface plasma waves during total reflection of light, resulting in a significant reduction in reflected light.When the wavelength of the incident light is fixed, the intensity of the reflected light is a function of the incident angle, and the incident angle corresponding to the lowest intensity of the reflected light is the resonance angle.The refractive index of the medium on the metal surface is very sensitive, and when the properties or adhesion of the surface medium change, the resonance angle will be different.
Simply put, SPR is based on the amount of ligands bound to the metal surface or whether they bind to generate different resonance angles.The change in resonance angle is proportional to the number of Ligand Analytes, in order to determine the binding state and Ka(thermodynamic affinity constant) of ligands with Analytes.
