Low T cell activation and polarization efficiency? An optimization guide for cytokine stimulation protocols
This article addresses the issue of low polarization efficiency in the in vitro activation and expansion of human T cells, systematically elaborating on the design of a co-stimulation protocol using anti-CD3 antibodies, anti-CD28 antibodies, and IL-2 under the "dual-signal + cytokine" three-signal model. It analyzes the effects of the concentration, coating method, and stimulation duration of each component on activation efficiency, and explores common influencing factors such as cell status, initial purity, and culture conditions, as well as their optimization pathways.
