This article focuses on the molecular characteristics and activation mechanisms of the NLRP3 inflammasome, systematically elaborating the role of the two-signal model (priming signal and activation signal) in NLRP3 inflammasome assembly and the maturation and secretion of IL-1β/IL-18, analyzing the differential regulation of phagocyte fate by different activation triggers and its impact on antitumor immunity, and exploring the pro-tumor mechanisms by which NLRP3 mediates angiogenesis, immunosuppression, and metastasis in the tumor microenvironment, as well as its tissue-specific exceptions.
