MAT1A protein
MAT1A protein: the core enzyme of hepatic methyl donor synthesis and a metabolic hub of epigenetic regulation
This article focuses on the molecular characteristics and biological functions of methionine adenosyltransferase 1A (MAT1A), systematically elaborating its enzymatic mechanism and structural basis as the primary producer of SAM catalyzing the synthesis of S-adenosylmethionine from methionine and ATP, analyzing the epigenetic consequences of formaldehyde exposure leading to reduced SAM content and loss of histone methylation through inhibition of MAT1A, and exploring the clinical significance of MAT1A silencing due to promoter hypermethylation in hepatocellular carcinoma and cholangiocarcinoma, as well as the therapeutic potential of restoring MAT1A expression or supplementing SAM.
