GPRC5D Protein: An Emerging Orphan Receptor Target for Immunotherapy in Multiple Myeloma
This article focuses on the molecular characteristics and immunotherapeutic target value of G protein-coupled receptor class C group 5 member D (GPRC5D), systematically elaborating its gene localization, structural composition, and homodimeric assembly as an orphan class C GPCR, analyzing its distribution characteristics of being confined to hair follicle regions in normal tissues while being highly specifically overexpressed on the surface of malignant plasma cells in multiple myeloma and its relationship of independent expression from BCMA, and exploring the preclinical and clinical activity of bispecific antibodies and CAR-T cell therapies targeting GPRC5D.
