DCAF16 protein
DCAF16 protein: an E3 ligase substrate receptor for covalent targeted degradation and a modular drug design platform
This article systematically elaborates on the molecular characteristics and biological functions of DCAF16, describing its structural localization and tissue expression features as a substrate receptor of the CUL4-DDB1 E3 ubiquitin ligase complex, analyzing its molecular mechanisms for recruiting novel substrates such as BRD4, SMARCA2/4, CDK4/6, and the androgen receptor through covalent modification of cysteine residues, and exploring its modular application value in the design of PROTACs and molecular glue degraders as well as its translational potential as a therapeutic target for triple-negative breast cancer.
