This article focuses on the molecular function of CDK4 and PROTAC-targeted degradation strategies, systematically elucidating the core mechanisms of the CDK4/6-cyclin D complex in regulating cell cycle progression and its aberrant activation characteristics in tumor cells. It analyzes the non-cell cycle functions of CDK4 in modulating tumor metabolism and anti-tumor immune responses, and explores the novel pathways provided by CRBN-based PROTAC technology for targeted degradation of CDK4.
